Guido Rodriguez scored the only goal of the match in the 13th minute, with a header, on a cross from Lionel Messi.
Argentina share the top of the group with Chile with 4 points in two games. Uruguay were starting out in the competition.
The two teams will now play again on Monday, Argentina against Paraguay and Uruguay against Chile.
Ten teams, divided into two groups, participate in the Copa America. The top four in each group advance to the quarter-finals.
