Am Institute of Clinical Pharmacology, Goethe University, Frankfurt (Director: Prof. Dr. Dr. Gerd Geisslinger), the position of a doctoral student is to be filled as part of a DFG-funded project. The project investigates the effect of fatty acid synthase (FASN) inhibitors as a potential therapy for glioblastoma (GBM). It is a collaboration between clinical pharmacology and experimental neurosurgery and neuro-oncology. The lipidome and metabolome are analyzed in plasma and tumor tissue from GBM patients without/with a preoperative ketogenic diet in order to identify the influence of certain lipid patterns that promote GBM growth. Using organotypic brain slice cultures with tumor transplants and GBM cell lines, in vitro mechanisms of FASN inhibitors alone and in combination with autophagy inhibitors are investigated. This includes lipid analyses, morphological-histological and molecular mechanisms. One focus here is the ability of glioblastoma cells to recycle lipids through lipophagy. Furthermore, the therapeutic effect of FASN inhibitors will be tested in vivo in an orthotopic mouse GBM model, including with regard to tumor growth, tolerability, pharmacokinetics, tumor morphology and GBM environment. Lipidome imaging will be established to display regional GBM lipid patterns in the GBM core and surroundings.
